Objectives:

  1. Relate protein structure to methods used to purify and characterize proteins.
  2. Relate protein structure to factors known to destabilize purified proteins (pH, temperature, degradative enzymes, adsorption to surfaces, oxidative damage).
  3. Describe four types of chromatography (ion exchange, hydrophobic, Gel filtration, Affinity) with reference to general principles of chromatography.
  4. Explain principle of electrophoresis in protein purification and determination of molecular weight (SDS-PAGE).
  5. Understand the principle of ultracentrifugation and how it is used to separate molecules and cellular structures.
